Effects of anti-platelet tubulin antibody on platelet functions were investigated. Monospecific antibody to human platelet tubulin was prepared in rabbits, which were given 4 injections of platelet tubulin isolated by 3 successive cycles of polymerization. The rabbit anti-tubulin serum gave a single precipitin line on double immunodiffusion against platelet tubulin and the high supernatant of a platelet sonicate (platelet extract). For studies on platelet functions, anti-tubulin F(ab')2 fragment was prepared from anti-tubulin rabbit serum. Platelets pretreated with anti-tubulin antibody showed decreased aggregation and 14C-serotonin release induced by collagen, but not by ADP or epinephrine. Platelet shape change, which was manifested by the decrease in light transmission after addition of collagen, was also inhibited by anti-tubulin antibody. 14C-serotonin uptake by platelets was significantly decreased in the presence of anti-tubulin antibody. Tubulin, possibly the membrane-bound one, may be involved in the mechanism of platelet aggregation and shape change induced by collagen.
