Extracts of cultures of E. coli carrying a plasmid with hepatitis B viral DNA contain a polypeptide that reacts with antibody to hepatitis B core antigen (anti-HBc). The antigen can be used in a radioimmunoassay for anti-HBc, either in microtiter plates coated with anti-HBc or by coating beads. The E. coli HBc antigen, liver antigen and a commercial assay gave very similar results in tests of 264 sera. The assay was also used to test 593 sera from patients attending a clinic for sexually-transmitted diseases.
