Intraneoplastic local injection of adriamycin for the treatment of experimental brain tumor--an autoradiographic study

Uptake and distribution of intraneoplastically administered adriamycin in brain tumors of rats was studied. The effectiveness of the chemotherapeutic drug by same method of administration for the treatment of the patients with malignant brain tumor was estimated. The tumor was induced in the brain of rats by intracerebral implantation of 1 .times. 104 Yoshida sarcoma cells (8 cases) or by transplacental administration of ethylnitrosourea (10 cases). Adriamycin (3H-ADM) was injected intraneoplastically into the brain tumor of rats. The rats were killed at various intervals (15 min, 1, 3 and 24 h, 7 days) and the tumors were studied immediately as to the distribution of 3H-ADM by light and electron microscopic autoradiography. 3H-ADM penetrated into the cells of neurinomas, oligo-astrocytomas and round cell sarcomas, 15 min after local administration. Diffuse uptake of 3H-ADM was found in the nuclei of many tumor cells especially in the area in contact with the CSF. The uptake of 3H-ADM was minute in the leptomeninges, ependymal layer, choroid plexus and cortex of cerebral base. In the surrounding areas of the tumor tissue there were no hemorrhage and necrosis of white matter. In EM autoradiographic studies, the uptake of 3H-ADM was clearly found in the chromatin of tumor nuclei. This chemotherapeutic agent is capable of direct penetration into the cells of an intracerebral tumor following intraneoplastic administration. Adriamycin apparently binds with the DNA in the nuclei of the tumor cells. Intraneoplastic local injection of adriamycin seems to be very effective for malignant brain tumor if it is used together with i.v. or intraarterial injection of the chemotherapeutic drugs.
