Study of polymorphous transformations of sulfanilamide during table manufacture

The possibilities of polymorphous transformations of the antibiotic sulfanilamide which take place during tablet manufacture were studied using differential calorimetric analysis. The study was carried out on the following: pure sufanilamide (before any processing), then after wet granulation (wetting by water), and then after compression at 120, 240 and 360 MPa, both on the initial product and on the granulated product; an equal parts mixture of sulfanilamide and dicalcium phosphate, before and after wet granulation on various granulators and after compression of the grains. Wet granulation or the wetting and compression provoke polymorphous transformations. A diluant (dicalcum phosphate) can to a certain degree protect the active ingredient from polymorphous transformations during wetting. Compression influences polymorphous transformations. Whatever the starting product, pure or diluted, wet or not, the differential colorimetric analysis curves of the tablets always show up the same transformations.
