A rapid radioimmunoassay was worked out for the determination of serum ethynylestradiol. After extraction with ether, the antiserum and the labeled antigen were added simultaneously, the mixture incubated first for 10 min on a 60.degree. C and next for 30 min on a 30.degree. C waterbath and after cooling the bound and free fractions separated on bone charcoal. The determination, including extraction, took < 5 h. The sensitivity of the method was 28.3 pmol/l, its internal variation coefficient was 4.53%, its external variation coefficient was 7.98%. The applied antiserum gave a 5.4% cross-reaction with estradiol-17.beta. and a 1.7% reaction with estrone; its reaction with other steroids was negligible.
