Preparation and cytochemical properties of Schiff-type dye-SO2 reagents: role of UV rays

A new method is reported for the preparation of Schiff-type dye-reagents, such as neutral red-SO2 and Bismarck brown-SO2. These dye-SO2 reagents were prepared with N HCl and sodium thiosulfate added to aqueous solutions of the 2 dyes. Neutral red-SO2 was shaken with 1.0 g and Bismarck brown-SO2 with 200 mg of activated charcoal/100 ml of the dye-reagents. Staining with neutral red-SO2 revealed excellent nuclear coloration in tissue sections [rat liver; kidney, intestine, and testes] which were hydrolyzed in HCl. Staining intensity with neutral red-SO2 at pH 4.0 was better than possible with the control at pH 2.6. Staining with Bismarck brown-SO2 at pH 2.5 without charcoal treatment revealed nuclei of orange color, whereas after charcoal treatment it revealed pale violet nuclei. The control used under the usual laboratory conditions exhibited less intense nuclear coloration when 0.5% Bismarck brown-SO2 at pH 2.6 was used. Staining with neutral red-SO2 at pH 2.6 when used under UV rays did not reveal any enhancement of staining intensity of the nuclei, whereas staining with Bismarck brown-SO2 at pH 2.6 under this condition exhibited definite increase in the intensity of nuclear coloration as compared with the control stained under the usual laboratory conditions. Apparently Bismarck brown-SO2 is more responsive to UV rays than neutral red-SO2. Possible interpretations of these findings are offered.
