Electrophysiological study of intravenous amiodarone in dogs

Adult mongrel dogs [6] were anesthetized with chloralose-urethane. With the histogram and programmed stimulation, the variations in the usual parameters of conduction and refractoriness of the A-V [atrio-ventricular] conduction system were analyzed, both before and after i.v. administration of 5 mg/kg of amiodarone (in a solution of Tween-80). No arrhythmias were produced. Evidently, amiodarone acts primarily at sinus and nodal level, to a lesser degree at atrial level, and not at all at the His-Purkinje level.
