A tumor-antigen (TA-4) of human squamous cell carcinoma and a radioimmunoassay for this antigen are demonstrated. TA-4, which has been purified from the cervical squamous cell carcinoma, is a glycoprotein with a molecular weight of approximately 48,000. Even though TA-4 activity was found in normal squamous epithelium of the female genital tract, clinical studies using radioimmunoassay revealed that the appearance of TA-4 in the circulation would be highly suggestive of the presence of squamous cell carcinoma (false-positive: less than 1%), and there was a nice correlation between the progress of the disease and the changes in serum TA-4 IeveIs. Attempts are now made to improve the sensitivity of the assay (55.2% of patients with cervical squamous cell carcinoma are positive at present). Serial determinations of serum TA-4 levels would be highly specific and useful for monitoring the disease.
