Induction of cellulase from Trichoderma reesei in defined culture media

Kinetics of induction of exo- and endoglucanase fractions of T. reesei QM 9414 cellulolytic complex was investigated, using a defined basal medium composed by glutamic acid 4 g/l, peptone 0.1 g/l and mineral salts. Lactose and cellobiose at the level of 0.1 and 0.5 g/l were added to the culture initially and at a time corresponding to 3/4 of the growth phase. Endoglucanase is induced by lactose and cellobiose, while exoglucanase is not, under the range of inducer concentration investigated. Both fractions are synthesized in response to the addition of low levels of native cellulose. The endoglucanase fraction is induced effectively by low concentrations of soluble disaccharides containing .beta. 1-4 linkages, while exoglucanase fraction required the presence of cellulose in the culture medium, the induction mechanism being probably triggered by intermediate oligosaccharides.
