Various preparative techniques for scanning electron microscopy were employed on specimens of normal kidney from an experimental animal and the advantages and disadvantages of each assessed with special reference to the information afforded in terms of renal glomerular architecture and the suitability of these methods for application to human needle biopsy material. Technically superior results were given by the cryo-fracture of fixed tissue followed by critical point drying. Critical point drying specimens which had been previously sectioned on a cryostat consistently exposed internal glomerular features. This latter method is of particular value where there is only limited tissue available (e.g., where small needle biopsy cores are dissected and cryostat sectioned for immunofluorescence or histochemical studies).
