Vascular responses to eserine in the frog (Rana tigrina)

The effect of eserine on perfused blood vessels of the frog (R. tigrina) was studied. Eserine produced a vasoconstrictor effect on different vascular beds. The mesenteric vasculature was more reactive than the systemic and pulmonary ones. This vasoconstrictor effect appeared to be a direct myogenic one and was not related to any cholinergic mechanism. Eserine in relatively higher doses potentiated acetylcholine-induced vasoconstriction and the effect was dose-dependent. The doses partially antagonized the vasoconstrictor effects of the ChE(cholinesterase)-resistant choline esters like carbachol, methacholine and bethanechol but the effect was more marked with lower doses. The potentiation of acetylcholine by eserine was marked during summers and negligible during winters. The antagonism by eserine of ChE-resistant choline esters was slight during summers but more marked during winters. The potentiating and antagonizing actions of ES apparently involved some cholinergic mechanisms.
