A modified Kohler technique of producing autogenous oral vaccine for swine

A one-litre flask containing 500 ml brain/heart infusion broth medium was inoculated with all the E. coli serotypes isolated from a particular outbreak, and incubated for 18-24 hours. It was then stored in a draught-proof isolated building. Calf replacer was added to water at twice the recommended dose and kept at 37 deg C. The bacterial culture was then poured into the calf replacer mixture and incubated for 8-10 hours. The mixture was stirred every two hours. Dry ground maize was added and absorbed most of the oral vaccine. This mixture was incubated for a further 10 hours. Acceptance of the vaccine was excellent and all strains were recovered from the maize on testing.
