Five day spleen cell cultures of 5 wk old (AKR/J.female. .times. BALB/c) or (BALB/c.female. .times. AKR/J)F1, but not parental AKR/J or BALB/c, mice spontaneously develop cytotoxic T Dd antigen, which is recognized by allogeneically induced anti-Dd CTL. The Kk antigen as judged by allogeneically induced CTL and the Kk-associated antigen as judged by spontaneously F1 CTL are present on 0.6, 2 and 4 day Con A-induced AKR/J targets. Cold target competition analyses show that unlabeled F1 cells prepared from 0.6 day in vitro incubation with Con A, but not from 2 or 4 days incubation, are able to block the cytotoxic activity of spontaneous F1 CTL on AKR/J targets. Early Con A-induced F1 targets do express a H-2Kk-associated antigen, which is recognized by these spontaneous F1 CTL. The Kk-associated antigen apparently is not identical to the MHC Kk antigen that is recognized by allogeneically induced anti-Kk CTL. The possible nature of these MHC associated antigens is discussed.
