The use of antibiotics in media for the isolation of fungi from polluted water

Sets of samples of sewage in various stages of treatment from the Dayton, Ohio, sewage treatment plant were surveyed to determine their mold populations. During parts of the survey two dyes and four broad spectrum antibiotics were compared as to their efficiency in developing these populations with a minimum of bacterial contamination. It was found that rose bengal was more effective than the closely related dye phloxine for this work. It was also found that chlortetracycline and streptomycin sulfate or dihydrostreptomycin sulfate could be held in aqueous solution over a period of at least three months under refrigeration during which time their effectivity did not appreciably decrease. Oxytetracycline did not decrease in effectivity over a two week period, but it apparently started to change before the end of that time. Chloramphenicol was not as effective as streptomycin in inhibiting bacteria possibly because it had a lower solubility under the conditions of the experiment. The medium recommended for enumerating the mold populations of polluted water and sewage is the basic Walkman soil mold plate count agar, non-acidified, in which phytone or soytone replaces peptone, and contains 0.035 gram per liter rose bengal and 35 μg. per ml. in aqueous solution of chlortetracycline. Oxytetracycline may be used if it is made up fresh for each experiment or if it is held in aqueous solution for only a few days.
