Rapid purification of chicken gizzard alpha-actinin

A method of purification was developed which yields highly purified .alpha.-actinin and requires less than 1 day to complete. The .alpha.-actinin is extracted from washed chicken gizzard muscle with water at 37.degree. Actin and a 55,000 dalton protein are quantitatively precipitated from the extract with 20 mM MgCl2. The .alpha.-actinin is subsequently precipitated from the extract by 30% (NH4)2SO4 and fractionated on DEAE cellulose. Spontaneous protein aggregation is prevented by adding 10% glycerol.
