Selective suppression by adherent cells, prostaglandin, and cyclic AMP analogues of blastogenesis induced by different mitogens

Depletion of adherent AMP analogs and an inhibitor of cAMP phosphodiesterase, methyl isobutyl xanthine, selectively suppress responses to the galactosyl-directed lectins. PGE1, cAMP analogs, and methyl isobutyl xanthine are most effective when added early in the course of lymphocyte activation. PGE1 levels are similar in the culture media of cells treated with the different mitogens, and are suppressed by indomethacin and the removal of adherent cells. Cells stimulated by different mitogens may differ markedly in their sensitivity to suppression by similar levels of PG. The graduation of sensitivity to PG and to cAMP of blastogenesis induced by the various mitogens studied, from most to least sensitive, is as follows: hepatic binding protein .gtoreq. peanut agglutinin > soybean agglutinin > Con A > PHA.
