Fluorimetric determination of ampicillin in calf serum

Sixteen calves (34-84 kg) were given ampicillin either by mouth (12 mg/kg) or i/m (4 or 10 mg/kg). After oral administration, serum concentrations were maximal after 2 hours (0.34 mu g/ml), falling steadily to 0.12 mu g/ml at 8 hours. After i/m injection of 4 mg/ml, the highest concentration was found after 30 minutes (6 mu g/ml), falling rapidly to <2 mu g/ml at 2 hours and 0.5 mu g/ml at 4 hours. After i/m injection of 10 mg/ml, the highest concentration was found at 1 hour (17 mu g/ml), falling to 7 mu g/ml (2 hours) and 2 mu g/ml (4 hours). An i/m dose of 4-10 mg/kg ampicillin is recommended, but to be fully effective should be given i/v thrice every 24 hours or twice in the case of s/c injection, to attain serum levels of 5 mu g/ml.
