Plasma concentrations of digoxin and digitoxin during digitalization of healthy dogs and dogs with cardiac failure

In normal and diseased dogs plasma concentrations of digoxin after high loading doses were similar to those after lower maintenance doses and it was concluded that the former were unnecessary in therapy. Digitoxin concentrations following a high loading dose varied considerably between animals and the usually recommended maintenance dose of 0.06 mg/kg/day produced low plasma values. The mean half-lives of digitoxin and digoxin were 13.1 and 31.3 hours respectively in healthy animals but, in some, unexpectedly short half-lives occurred. The importance of administering the total daily dose in several parts is emphasized.
