On the origin of leukocyte procoagulant activity

Data support the argument that blood mononuclear cells, containing less than 1% granulocytes, are capable of generating tissue factor [TF] activity when stimulated in vitro by antigens, mitogens, and endotoxin. Two patients with monocytic leukemia were also studied in order to address the importance of cell maturity and the effects of chemotherapy on monocytes. Immature monocytes or monocytes previously exposed to cytotoxic drugs may have an impaired ability to generate TF activity. Physical methods for removal of the cells from the plates or endotoxin contamination of the reagents may account for spontaneous TF generation sometimes observed in unstimulated cultures.
