Studies on the shelf life of Schiff's reagent prepared with different organic acids

Schiff's reagent was prepared with citric, tartaric or oxalic acid in place of the usual 6 N HC1. When acid hydrolyzed tissue sections were stained with these dye-SO2 reagents, more intense staining occurred with citric or tartaric acid-Schiff than with conventional Schiff's reagent or one prepared with oxalic acid. Schiff's reagent prepared with citric or tartaric acid retained its staining ability much longer than the reagents prepared with 6 N HC1 or oxalic acid. The same facts hold true when PAS[periodic acid-Schiff]-positive substances are stained with these newly modified Schiff's reagents. A possible explanation for increase in staining intensity of nuclei or of PAS-positive substances with tartaric acid-Schiff and citric acid-Schiff is offered.
