Staining of DNA with aqueous solution of basic fuchsin after phosphoric acid extraction of RNA

A method is described for staining tissue sections using an aqueous solution of basic fuchsin following extraction of RNA with cold concentrated phosphoric acid for 20 min or in 75% acid for 2 h. Optimum staining is possible at pH 4 and 5.5. The peak of maximum absorption of stained rat nuclei is 560 nm, but that of an aqueous solution of basic fuchsin is 540 nm. The mechanism of staining may be binding of DNA-phosphate groups with the dye.
