Immobilized asparaginase and its use in medicine

Various methods of asparaginase immobilization and the biochemical characteristics of immobilized preparations were discussed in this literature review. Physical adsorption, covalent bonding, gel-inclusion and micro-encapsulation were the methods applied to asparaginase from Bacillus cadaveris [Clostridium cadaveris], Escherichia coli, Pseudomonas fluorescens, Erwinia carotovora and Proteus vulgaris. Results were discussed from various tests of biological activity of immobilized asparaginase in humans, mice, rats and monkeys. Data demonstrated the efficacy of immobilized asparaginase in producing almost no antibodies to asparaginase in treating leukemia. Asparagine concentration in the blood significantly decreases with administrations of the enzyme in microcapsule or granule form. The most promising results were obtained with administrations of extracorporeal asparaginase reagents which have demonstrated the possible avoidance of fibrose deposit formation.
