There are concerns that .alpha.-fetoprotein, a high-affinity estrogen binder in rat uterus, may mimic estrogen receptors in various human neoplasms, Using a sucrose gradient method and a dextran-coated charcoal method, human .alpha.-fetoprotein from germinal cell cancers did not have detectable estrogen-binding activity or interfere with estrogen receptor assay. Estrogen receptors detected in various human cancers by sucrose gradient and dextran-coated charcoal methods are not .alpha.-fetoprotein in origin.
