Isoprenaline relaxation in vascular smooth muscle of aged rats. Effect of flunarizine

Aorta rings depolarized in the presence of phentolamine 2 .times. 10-6 M showed a reduced relaxation when the animal aged. Some preparations obtained from 30 mo. old rats were insensitive to isoprenaline. Tachyphylaxis was observed with rats 6 mo. old or older. When the aortae were incubated for 90 min in the presence of flunarizine 10-9 M, tachyphylaxis was reduced. This flunarizine action might be related to a reduction of the desensitization of the .beta.-adrenergic receptor occurring in old rats, and not to Ca influx inhibition.
