A simple and rapid test for the assay of Co-activated acylase activity in human serum and tissue homogenates is described. The test was based on use of dry paper discs soaked with reagents necessary to perform the enzyme reaction. Mean serum enzyme activity on the 1st day of jaundice in adult patients with hepatitis was 22.1 .+-. 13.8 mU/ml, and in youngsters 8.7 .+-. 6.7 mU/ml. No acylase activity was found in urine. Enzyme activity appears in serum at least several days before onset of jaundice; this test can be applied for early diagnosis of viral hepatitis.
