Lymphocyte and serum-mediated immunity in murine osteosarcoma

The time-sequence relationships of lymphocyte cell-mediated immunity (CMI), antibody-dependent cellular cytotoxicity (ADCC) and serum blocking activity (BA) were tested in mice injected with Dunn osteosarcoma cells. CMI was present but weak on the 1st day after tumor implantation. It peaked on the 8th day and then diminished, although it was detectable for the duration of the experiment (17 days). The ADCC assay was positive on the 1st day and peaked on the 5th day. Beyond 5 days there was no significant ADCC. Serum BA was not significant until the 5th day but remained active for the duration of the experiment. An assay to determine the site of BA was conducted on the 8th day, the peak of BA. Preincubation of lymphocytes with serum did not affect their killing capacity, while preincubation of the tumor cells resulted in BA of 30%.
