The localization of hepatitis B surface [Hbs] and core [Hbc] antigens in the hepatocytes was studied in liver specimens taken from 7 patients with Hbs antigenemia and from 1 control subject, by indirect immunofluorescence, EM and indirect immunoelectron microscopy with peroxidase. A cytoplasmic fluorescence was demonstrated with anti-Hbs antibodies and a nuclear fluorescence was demonstrated with anti-Hbc antibodies. Tubular formations were found in the dilated cisternae of the smooth endoplasmic reticulum and 20 nm spherical particles were found in the nuclei of hepatocytes. Immunoperoxidase, showed that the intracisternal tubular formations support Hbs antigen and the intranuclear particles support Hbc antigen.
