The determination of disaccharides resulting from digestion with chondroitinases is a very useful means for the identification of the components present. Paper chromatography is applicable to amounts of disaccharides ranging from 25-100 .mu.g of each isomer; however, the procedure requires about 36 h. A more rapid and sensitive method for the qualitative and quantitative determination of these disaccharides is described. Cellulose TLC plates give satisfactory separations and detection of amounts of disaccharides as low as 2.5 .mu.g. The application of color reactions with the eluted spots allows their quantification. An advantage of this method is the short period of time (12 h) required for the development of the TLC chromatograms.
