Effect of prostaglandins on gastrointestinal smooth muscle

The possibility of a relationship between prostaglandin (PG) and 5-hydroxytryptamine (5-HT) with regard to gastrointestinal motility was investigated. The studies were conducted in vivo in albino mice and fecal output was taken as the index of gastrointestinal motility. In vitro studies were conducted on isolated guinea pig ileum and rat fundus. 5-HT receptor antagonists (bromolysergic acid diethylamide, methysergide, BW 501C67 [.alpha.-anilino-N-2-m-chlorphenoxy-propylacetamidine] and cyproheptadine) and PG receptor antagonist (SC19220 [1-acetyl-2-(8-chloro-10,11-dihydrodibenz[b,f] [1,4]-oxazepine-1-carbonyl)hydrazine]) reduce PGE2 and 5-HT induced increased motility in vivo and increased spasm in isolated preparations in a similar manner. Acetylcholine induced spasms were not affected by either of the antagonists in vitro. A possible relationship between PGs and 5-HT in the smooth muscle of gastrointestinal tract was suggested.
