The fermentation of after-yeast molasses stillage was studied by separating the process of methane fermentation into 2 phases: acid and alkaline. Vitamin B12 is formed chiefly in the 2nd phase. A change in the main biochemical indices and accumulation of vitamin B12 were affected by inactivation of the phase I culture and the different rate of medium flow into the apparatus of phases I and II. The fermentation of after-yeast molasses should be separated into 2 phases as it enables a uniform supply of nutrient medium to the apparatus, prevents over-dosage, increases the vitamin B12 yield and decreases the biological O2 demand of the waste, which is important in applying the process for biochemical purification.
