The results of these interesting studies support a benefit for rituximab therapy after initial treatment, but do not resolve the question of whether there is a benefit for regularly scheduled re-treatment versus rituximab re-treatment at the time of disease progression. Hainsworth et al. reported at American Society of Hematology (ASH) 2003 a study of 114 patients randomized to maintenance rituximab weekly × 4 every 6 months for 2 years versus re-treatment at progression. While the median PFS was longer on the maintenance arm (31 vs 8 mo), there was no difference in the median duration of rituximab benefit (31 vs 27 mo). Although the latter finding suggests a benefit for either strategy, this study did not have sufficient power to detect a difference.
