Histochemical examination of post-embryonic, migrating and muscle larvae and intestinal trichinae of Trichinella spiralis revealed lipid droplets distributed throughout the body of post-embryonic, migrating and young muscle larvae. In older muscle larvae and trichinae, lipid was found particularly in the stichocytes, and in blastulating embryos in trichinae. Older muscle larvae accumulate glycogen and other PAS-positive substances which reach a maximum 25 days after infection of the host. Glycogen is rapidly depleted in intestinal worms, in which mucopolysaccharides were found in the stichocytes, hypodermal carinae, and reproductive cells of the males. The cytoplasm of post-embryonic larvae was pyroninophilic. Pyroninophilia increased in cells of young muscle larvae especially the stichocytes, later decreasing in the hypodermis, muscle cells and reproductive cells but remaining high in the stichocytes, which are the only positive sites in intestinal worms. Blastulae and post-embryonated larvae give a strong positive Feulgen reaction which takes the form of coarse grains but little reaction was seen in muscle larvae and trichinae.
