Comparative study of the resorptive and excretory properties of some repellents

Solutions of 8 repellents in 70% alcohol were applied to the skin of rabbits, and the rates of absorption into the blood and excretion in the urine were determined. The 8 repellents were deet, DEAVK [N,N-diethylpentanamide], R-2 [N,N-diethylbenzamide], R-203 [N,N-diethyl-2-phenoxyacetamide], PVK [1-(1-oxopentyl)piperidine], R-162 [1-benzoylpiperidine], R-228 [1-(3-methylbenzoyl)piperidine] and PFUK [1-(phenoxyacetyl)piperidine]. Absorption rates were found to be proportional to the partition coefficients of the compounds between oil and water, the more lipid-soluble compounds being absorbed more rapidly. The average blood concentrations during the first 8 h after application of 40% solutions at 1.2 ml/kg body weight ranged from 22.41 mu g/ml for deet to 146.46 for R-228. Excretion in the urine was found to depend partly on molecular weight and partly on solubility in water, the smaller and more soluble compounds being excreted more rapidly. During the first 8 h after application of the solutions, urine concentrations expressed as a percentage of the blood concentration ranged from 2.66 for R-203 to 54.32 for PVK. It is suggested that rates of absorption and excretion should be taken into account in studies on the toxicity of repellents and in choosing the least toxic for practical use.
