Isolation of mitochondria from mouse mammary gland

When isolating mouse mammary gland mitochondria it is essential to (i) remove as much milk from the mammary tissue as possible and (ii) use protective agents, such as bovine serum albumin, to absorb the free fatty acids before attempting to homogenize the tissue. The techniques used for (i) and (ii) in different stages of tissue development and the centrifugation process are described in detail. The mammary mitochondria obtained by this process are coupled, intact and demonstrate energized ion transport.
