The interaction between pentagastrin and cholecystokinin on pancreatic secretion in man

Three healthy men, 23 years of age and weighing 70 to 78 kg, had double-lumen Lagerlof tubes passed after at least 8 h fasting. Intravenous infusions of saline, cholecystokinin or pentagastrin, or cholecystokinin with pentagastrin, were given during six 15-min periods. Polyethylene glycol (PEG) was infused as a non-absorbable marker. Recovery of PEG averaged 69% and showed notable variations. About 2.4% of the PEG infused into the duodenum was recovered from the stomach. Recovery of PEG from the duodenum during infusion of saline was 60, of pentagastrin alone 56%, of cholecystokinin alone 74, and of cholecystokinin and pentagastrin 73%. The secretion of bicarbonate, proteolytic enzymes and bilirubin was slightly greater in response to pentagastrin than to physiological saline, but no dose-dependent effect was observed. Cholecystokinase showed a dose-dependent effect on bicarbonate, proteolytic enzyme and bilirubin secretion; the response being greater at the lowest dose of cholecystokinin than to saline. Pentagastrin showed no effect on cholecystokinin-induced secretion of bicarbonate. The enzyme and bilirubin responses to cholecystokinin with pentagastrin were variable: in 2 of 3 men no effect was observed, but in the third pentagastrin increased the initial enzyme and bilirubin responses to cholecystokinin. A positive correlation was found between proteolytic enzymes and bilirubin in the duodenal content.
