The stability of normal and hyperacetylated chromatin was studied in cell-free system of Drosophila embryos. Structural rearrangement of chromatin took place in this system, resulting in the release of a portion of nucleosomal DNA supercoils and increased chromatin sensitivity to DNAse. This rearrangement also supported a high level of nucleosomal DNA conformational mobility. It was shown that the structural rearrangement is much more effective in cases of hyperacetylated chromatin.
