Storage of bluetongue virus-infected Culicoides variipennis

Colonized female Culicoides variipennis were orally infected by blood feeding on a sheep with bluetongue viraemia. These flies had a subsequent infection rate of 63%. Batches of flies were stored for up to 2 years under 8 different conditions. This insect-source virus was highly stable when stored at 4 deg C in a phosphate-buffered saline-antibiotic solution containing 0.5% bovine serum albumin as a protein stabilizer. The virus was considerably less stable when stored at -100 deg or -20 deg C. The usual standard blood-virus anticoagulant virus-preservative solution (consisting of potassium oxalate, 5 g; phenol, 5 g; glycerin, 500 ml; and distilled water, 500 ml) was highly detrimental to storage of the insect-source virus.
