Experimental animal studies and clinical trials have demonstrated the efficacy of losartan, an active antihypertensive agent. It proved to lower systolic and diastolic pressure (by the average of 21 mm of mercury column), wedge pressure in the pulmonary capillaries and total peripheral vascular resistance. Losartan also showed positive effect on the left ventricle morphology by reducing the thickness of the posterior wall from 10.1 to 9.9 mm, and that of the interventricular septum from 10.2 to 1.0 mm. The drug demonstrated good tolerance and higher efficacy compared to angiotensin-converting enzyme inhibitors of the first generation (captopril) and second generation (enalapril). Antihypertensive effect of losartan in the dose of 50 mg per day is comparable to that of 5 to 10 mg felodipine after 12 weeks of therapy.
