The sample, 2 ml serum, 1 g tissue, 0.25 g dried grass or meal, was heated with 3 ml HNO3 and HClO4 (4: 1), 2 small glass balls and a drop of octan-1-ol in a metal block at about 100oC for 30 to 45 min, at about 210 degrees for about 45 min and heating was reduced when the volume was about 0.5 ml.
