Beneficial effect of 1,25-dihydroxyvitamin D3 in the treatment of myelofibrosis

In a man, 62 years old, with chronic idiopathic myelofibrosis unresponsive to treatment with oxymetholone, 1,25-dihydroxycholecalciferol 2.5 micro g daily for 6 months and a diet low in energy associated with corticoids from the 4th month resulted in a normalization of blood values and improved bone biopsies. Advanced myelofibrosis and osteomyelosclerosis were replaced by hyperplasic panmyelosis comparable with the initial or cellular stages of myelofibrosis. The same dose of 1,25-dihydroxycholecalciferol in a woman, 72 years old, with acute myelofibrosis or megakaryoblastic leukaemia resulted in a normalization of blood values without recourse to blood transfusions. After a worsening of her condition after 5 months treatment the patient showed remission on the same treatment and remained in remission during maintenance therapy.
