A new method for the sequential extraction of oxytocin and antidiuretic hormone (ADH) from plasma for radioimmunoassay is reported. The procedure involves the adsorption of these peptides on Fuller's earth and selective and sequential elution of oxytocin using aqueous acetone, and ADH with ammonia-ethanol mixture. This method is simple, reproducible and is useful for processing of a large numer of samples simultaneously. The extraction and separation of both oxytocin and ADH from the same sample would be helpful in performing many investigations on the levels of neurohypophyseal hormones in physiological and pathological conditions in the biological fluids.
