Genotype study of T receptors in the diagnosis and classification of leukemias and lymphomas

Using the restriction enzymes EcoRI and Hind III and a probe that detects the constant beta regions of the T receptor genes, we have carried out genotype studies in 41 patients with different types of leukemias and lymphomas. The aim of the study was to evaluate the sensitivity and the specificity of these methodologies for the characterization of T lymphoproliferative syndromes. Clonal rearrangements were found in: 7/8 T lymphoblastic leukemias; 1/11 B lymphoblastic leukemias; 2/6 chronic T lymphocytosis and in the only chronic T lymphatic leukemia that was studied. Clonal rearrangements were not observed in chronic B series lymphoproliferative syndromes (9 chronic lymphatic leukemias and 2 lymphomas) nor in 2 Hodgkin's lymphomas. None of the 20 normal controls showed restriction polymorphisms. These results confirm that genotype studies can be highly useful, along with cytology and immunohistochemistry, for the categorization of T-lymphoproliferative syndromes.
