The effect of single dose administration of compound 48/80, a potent histamine releaser, on the pathogenicity of E. histolytica was studied in four isolates grown and maintained in modified Boeck and Drbohlav's medium. The caecal pathogenicity of the isolates was assessed in just weaned rats. Groups of infected rats were given a single dose of compound 48/80 ip on days 1, 2, 3, 4 or 5 post-infection. The caecal pathogenicity scores were significantly increased in groups that received compound 48/80 on days 3, 4 and 5 post-infection. The maximum enhancement of pathogenicity was recorded in animals that received the compound on day 3. It is suggested that raised peripheral level of histamine may act as a trigger to augment the virulence of E. histolytica after a period necessary for colonization.
