Analysis of the c-fms gene in hematopoietic neoplasia with changes of chromosome 5

We analyzed for c-fms DNA levels in hematopoietic neoplastic cells with 5q-or-5 abnormalities. Lymphoid leukemia cell lines (NALM 6 and PEER) with 5q-changes had no c-fms rearangement or deletion. On the other hand, DNA obtained from a female case of 5q-syndrome had an approximately half intensity hybridization with the v-fms probe, indicating the 5q-cells deleted the c-fms gene. DNA from myelodysplastic syndrome with -5 change, as well as other numerical and structural changes, also had a deletion of the c-fms gene. In the lymphoid leukemia cells, NALM 6 had a del (5) (q32), whereas PEER showed del(5)(q22q31). Thus, the c-fms gene might localize at 5q31.fwdarw.q32 region, which is a crucial cytogenetic region of 5q-syndrome (5q-23.fwdarw.q32). These findings suggest that the c .cntdot. fms gene is deleted in the cells from 5q-syndrome, and the deletion might be involved in the pathogenesis of this disorder.
