Treatment of myelodysplastic syndromes with danazol

42 patients with myelodysplastic syndromes (MDS) were treated with 600 mg of Danazol, for a minimum of three months. 10 patients showed a good response and 6 had a partial response, whereas the 26 remaining were considered a failure. The survival median of the patients with a positive response was 43 months, which is significantly better than the 7 months which were observed in those patients in which this form of treatment was considered a failure (p V 0.01). The observed toxicity was minimal and no therapeutic modifications were required. The morphological subgroups with refractory anemia and refractory anemia with an excess of blast showed the best responses. The use of Danazol in patients with MDS should be considered an option, since it has minimal toxicity and can significantly modiy hematologic parameters and survival. This drug might be used in initial therapy, before other more toxic ones are employed in MDS.
