A method for comparative and long-term studies of drug absorption from selected portions of the gastro-intestinal tract in normal dogs is described. The method involves the use of two or three cannulas implanted in specific locations of duodenum, jejunum, or ileum or in all three parts of the small intestine. Thus these areas plus the oral and rectal routes are made accessible for depositing compounds precisely at selected absorption sites within the gastro-intestinal tract. The dogs may be used repeatedly for comparative evaluation of drugs or formulations, as well as for determination of absorption patterns. A comparative study with 16 beta-methylprednisone and its 21-phosphate ester was used to demonstrate utilization of this method.
