Generation of cytotoxic T cells to cytomegalovirus in patients with cervical intraepithelial neoplasia

Peripheral blood lymphocytes (PBL) from patients with cervical intraepithelial neoplasia were stimulated with cytomegalovirus (CMV) antigens and analysed for function and phenotype over a period of 14 days in culture. Monoclonal antibodies OKT4, OKT8 and Leu7 were used to identify T cell subsets and these were analysed on a flow cytometer. Cytotoxicity assays were performed on autologous non-infected and CMV-infected cervical (Cx) cells. In one of three patients CMV-specific and HLA-restricted cell lysis was demonstrated. No evidence was obtained of expression of CMV antigens on non-infected Cx cells grown from these patients. NK cell lysis was evident in uncultured PBL in assays against autologous Cx cells and K562 target cells, but this activity was lost on culture.
