Effects of bromobenzene, thioacetamide and carbon tetrachloride on activities of alcohol dehydrogenase and aldehyde dehydrogenase of hepatic subcellular fractions in C57BL and DBA mice

Effects of bromobenzene, thioacetamide and carbon tetrachloride (CCl4) on the activities of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in C57BL/6J and DBA/2 strain mice were studied. 1) The effect of bromobenzene treatment on ADH and ALDH activities was different between C57BL mice and DBA mice. Activities of ADH, high Km- and low Km- ALDHs in the soluble fraction in C57BL mice were not affected by this drug, however, these activities decreased in DBA mice. The activities of high Km- and low Km-ALDHs in the mitochondrial fraction and high Km-ALDH in the microsomal fraction decreased in C57BL mice. While the high Km-ALDH activities in the mitochondrial and microsomal fractions were not affected in DBA mice, the mitochondrial low Km-ALDH activity decreased as in C57BL mice. 2) The effect of thioacetamide treatment on ADH and ALDH activities of C57BL mice was similar to that of DBA mice except for the activity of microsomal high Km-ALDH. Activities of ADH and high Km-ALDHs in the soluble and mitochondrial fractions remained unchanged while activities of low Km-ALDHs in the soluble and mitochondrial fractions were decreased. Microsomal ALDH activity in C57BL mice decreased by this drug but it remained unchanged in DBA mice. 3) There was a similarity in the effect of CCl4 treatment on ADH and ALDH activities between C57BL and DBA strain mice except for that activity of cytosolic low Km-ALDH. Activities of ADH and cytosolic high Km-ALDH were elevated after CCl4 treatment while the activities of high Km- and low Km-ALDHs in the mitochondrial fraction and microsomal high Km-ALDH decreased. Cytosolic low Km-ALDH activity was elevated by CCl4 in C57BL mice but it remained unchanged in DBA mice.
