Effect of dextran sulfate on the growth of cultured fibroblasts derived from normal human skin and keloid lesions

The in vitro effect of dextran sulfate on the growth of fibroblasts was examined in 4-day fibroblast cultures, including four normal human foreskins and two keloids. The results revealed that dextran sulfate at low concentration (100-1,000 .mu.g/ml) stimulated thymidine incorporation, but inhibited fibroblast growth at a higher concentration (.gtoreq. 10 mg/ml). The inhibitory effect of dextran sulfate was due to its direct cytotoxic effect rather than its high osmolality, interference with cellular attachment or its combining with essential serum growth factors.
