Determination of sophoramine and sophocarpine in rabbit plasma by capillary column gas chromatography and their pharmacokinetics

A rapid, sensitive and specific method for the determination of sophoramine in rabbit plasma by capillary column gas chromatography was proposed. The recovery of sophoramine from plasma was over 90%. There was a linear relationship between 0.5 and 10 .mu.g. The comparative pharmacokinetics of sophoramine and sophocarpine (iv 10 mg/kg) were studied in rabbits. Plasma concentration vs time data were described by an open two compartment model. The pharmacokinetic parameters of sophoramine and sophocarpine were as follows: Vc 0.72 and 1.40 L/kg; CL 17.0 and 28.2 ml/(kg .cntdot. min-1); .alpha. 0.126 and 0.114 min-1; .beta. 0.0136 and 0.00853 min-1, respectively. The differences in distribution volume, clearance and elimination rate constant of the drugs were significant (p < 0.05).
